ISOBOLAR ANALYSIS OF THE BINARY FIXED-RATIO COMBINATION OF ACETYLSALICILIC ACID-ACETAMINOPHEN

The present study is part of a series of larger investigations concerning combinations of paracetamol with several other drug substances, as a rational modality of pain therapy. By using a nociception model with chemical stimulus (Zymosan A) and the administration of fixed-ratio dose sequences of paracetamol and acetylsalicylic acid we attempt the identification of the type of pharmacodynamic interaction between those two substances. The evaluation of the nociceptive response is quantal, while for the evaluation of the interaction we have used the isobolar analysis (composite additive line method). The data obtained suggested a synergistic interaction, demonstrated by the following parameters: Zadd = 253.16 ± 33.90 mg/kg body weight (b.w.), Zmix = 75.00 ± 19.02 mg/kg b.w, Zmix < Zadd, γ = 0.296 (Ft = 4.460, Fc = 14.414, Tt = 3.925, Tc = 4.275 p < 0.01).
