Orthodontic conditions represent a serious public health issue, affecting patients of all ages. The current treatment of these conditions implies the utilization of clear aligners (CAs) that improve teeth alignment within the periodontium. The safety and biocompatibility of these dental products used in orthodontics is essential for their use in therapy. Thus, the aim of the present study is to assess the cytocompatibility and safety profile, in vitro (using HaCaT keratinocytes and HGF fibroblasts as experimental models) and in vivo (on the chorioallantoic membrane) of two CAs types (CA1 and CA2) used in orthodontic treatments. The results indicated that the 24 hours exposure to CA1 and CA2 induced no cytotoxicity in HaCaT and HGF cells, the cell viability percentages maintaining over 70%. CA1 and CA2 caused no significant impact on cell morphology, apart from a loss of confluence observed at the dilution of 1:1. Moreover, CA1 and CA2 induced no toxicity at the vasculature of the chorioallantoic membrane, being categorised as non-irritant.
