THE THERAPEUTIC EFFECT OF HUANGLIAN JIEDU DECOCTION ON ALZHEIMER’S DISEASE BY REGULATING PHAGOCYTOSIS INDUCED BY Aβ IN BV-2 MICROGLIAL CELLS

This study aimed to investigate the effect of Huanglian Jiedu Decoction on amyloid beta (Aβ) phagocytosis in BV-2 microglial cells and its regulation on Trem2 protein expression and secretion. BV-2 cells were cultured with different doses of Huanglian Jiedu Decoction. The Aβ phagocytosis of BV-2 cells was determined by flow cytometry. Western blot was used to detect the Trem2 protein expression in BV-2 cells. The Aβ phagocytosis in BV-2 cells after Trem2 knockout was compared. The results showed that 10 μM and 20 μM Huanglian Jiedu Decoction significantly promoted the Aβ phagocytosis of BV-2 cells compared with the control group. The expression of Trem2 protein in BV-2 cells in 5 and 10 μM Huanglian Jiedu Decoction groups was slightly increased compared with the control group, but without reaching the statistical significance. The expression of Trem2 protein in 20 μM dose group was significantly increased compared with control group. In conclusion, Huanglian Jiedu Decoction can promote the Aβ phagocytosis in BV-2 cells by upregulating the expression of Trem2 protein expression in a dose-dependent manner.
