In the complex, well-coordinated process of inflammation, a variety of cell types and molecules cooperate in a cascading network. Numerous physiological and pathological processes are rooted in inflammation. The mechanism of inflammation comprises a number of dynamic, coordinated reactions, including cellular and vascular events and specific humoral secretions. The liver is adversely affected by proinflammatory cytokines such as interleukin-1β (IL-1β), IL-6, and tumour necrosis factor. IL-6 has been shown to selectively regulate several cytochrome P450 (CYP) enzymes, including CYP3A4. Some CYPs actively contribute to inflammation by converting fatty acids into mediators that are either pro- or anti-inflammatory. Genetic variability within inflammatory transcription factors is of great interest due to its critical role in inflammatory signalling.
