Abstract:
Chemotherapy-induced nausea and vomiting (CINV), is one of the most common adverse reactions to chemotherapy and the
use of preventive medication can influence the success of the therapy. The aim of this study was to assess in vitro and in vivo
the effect of olanzapine on the invasion and metastasis of lung cancer cells and the prevention of nausea and vomiting after
chemotherapy. The invasion and metastasis of lung squamous carcinoma cells was evaluated in vitro on A549 cells and in
vivo on a nude mice non-obese diabetic (NOD)/severe combined immunodeficiency (SCID) lung metastasis model. Also
thirty eight lung cancer patients who received chemotherapy were included in the study. The preventive effect of olanzapine
on chemotherapy-induced nausea and vomiting was analysed. In vitro experiments showed that olanzapine decreases the cell
number and rate of migration and invasion of A549 cells compared to control groups. In vivo olanzapine decreased the
metastasis diameter in olanzapine group compared with the control group. The use of olanzapine for preventing
chemotherapy-induced nausea and vomiting in lung cancer patients has the benefit of also inhibiting the invasion and
metastasis of lung cancer cells.
