Abstract:

The objective of the study was to evaluate the relative influence of setting parameters in dry granulation (compaction) and tableting phases on physical characteristics of clarithromycin tablets: disintegration time, hardness, weight variability and friability. Full factorial experimental design was applied. For compaction phase the following variables have been considered: compaction force, gap, role speed and sieve, while for tableting phase, compression force and tableting speed were evaluated.
As a result, we have evaluated the design space in what concerns the parameters evaluated and the relative influence of each variable on the desired characteristics of the granulate and tablets. We were able to establish optimal parameters setting to assure the highest productivity and compliance of the product with the standard specifications.
