THE INFLUENCE OF PHYTOHORMONES FOR IN VITRO CULTURE OF SOME CHLOROPHYTA FROM THE BLACK SEA COAST.

Abstract:
A great number of specific compounds of algal origin have been purified and characterized and are being used experimentally in medicine. In order to obtain sterile algal biomass, the aim of our studies was to establish the optimum condition for in vitro cultivation, for the future extraction of useful algal compounds. We used young thalli of three species of Chlorophyta: Ulva rigida, Enteromorpha intestinalis and Cladophora vagabunda. The inoculated fragments were differentially drawn from the three zones of the thalli: basal, median and apical, measuring 1 cm each. The sterilization was made with lactic acid and the cultures were made in three different nutritive mediums: Provasoli artificial medium (ASP2), Von Stosch, enriched Provasoli sea water (PES), which were supplied with phytohormones: indole-3-acetic acid (IAA), gibberellic acid (GA), kinetin (K), in a number of experimental variants.
The microscopic and macroscopic observations and also the gravimetric measurements we made, allowed us to determine the rate of growth of the tested sea algae and finally, the elaboration of an optimum cultivation protocol for the analyzed algal species. We also establish the optimum concentrations and the positive phytohormones ratio. We consider that the present studies are a really start point for the future studies in order to obtain pharmaceutical algal compounds and for finding the cell mechanisms by means of which the phytohormones influence the morphogenesis in the case of green, pluricellular sea algae.
