PREPARATION AND CHARACTERIZATION OF MICROEMULSIONS WITH HYDROCORTISONE ACETATE.

Abstract:

The aim of this study was to formulate and to characterize a topical microemulsion with hydrocortisone acetate (HA). The excipient selection was made according to the literature data as follows: isopropyl miristate was selected as oily phase, distilled water as aqueous phase, polysorbate 80 as surfactant and propylene glycol as cosurfactant. The ratios of each component are obtained from several investigations : the study of the solubility of HA in the surfactant and cosurfactant in order to select the ratio between the surfactant/cosurfactant; a phase diagram was constructed in order to determine the proportion of the three phases (oily phase, watery phase and the mix of the surfactant and cosurfactant).
Stability studies were performed at several temperatures (40C, 250C, 400C) for a selected formula of microemulsion. A reduction of viscosity was observed for all temperature values and a slight modification of pH values. All other parameters tested (appearance, centrifugation test, emulsion type) remained practically unmodified during the study at the considered temperatures.
