FORMULATION AND EVALUATION OF A PREVENTIVE ANTIDOTE IN ORGANOPHOSPHOROUS POISONING .

Abstract:

Due to the large utilisation of organophosphorous pesticides in agriculture in almost all countries there are many units for their production, analysis or storage as well as research institutes. Following different accidents, including terrorist attacks, massive contamination with toxic organophosphorous derivates may appear. Since the main toxic symptoms concerns cholinestarase inhibition, pyridostigmine tablets are useful as preventive antidote in such intoxication. The restriction in providing large quantities of such antidote appears from its low stability following water adsorption from athmosphere. Very stable tablets were prepared using large quantities of bentonite as excipients. Since pyridostigmine proved a strong adsorption on bentonite in vitro, in vivo behaviour was tested by evaluation of its pharmacokinetics at 4 healthy volunteers. It was found that in vivo absorption was less influenced as was expected after in vitro experiments.
