Abstract:

Chlorthalidone is a practically insoluble in water diuretic, used for the treatment of edema and hypertension. β-cyclodextrin was used as a host molecule to improve its solubility in water, via inclusion complex formation. In order to obtain the inclusion complexes, different molar ratios (1:1, 1:2) and various preparation methods (physical mixing, kneading, spray-drying) were used. Phase solubility diagram of chlorthalidone and β-cyclodextrin was constructed and a diagram of AL type was obtained, with a stability constant of 248 M-1. The inclusion complexes were analysed by X-ray diffraction and 13C NMR techniques. As the solubility in water increased, the partition coefficients of the products decreased.
