PREPARATION AND IN VITRO CHARACTERIZATION OF COLON RELEASE PELLETS WITH pH AND TIME CONTROL MECHANISM.

Abstract:

The aim of this experimental work was the preparation and in vitro study and characterization of some pellets with colonic delivery due to pH and time-control.
The pharmaceutical system contains a retardation layer (hydroxypropylmethyl cellulose and Eudragit RS) and an enteric layer (Eudragit L 55). The most important formulation factors for colonic delivery are: the amount of HPMC that expand a sufficient pressure to break Eudagit RS polymeric layer and the optimum ratio amongst thickness of Eudragit RS and amount of plasticizers that allow to obtain a flexible polymeric film that resist 3-4 h in 6.8 pH-medium and then break under the force exercised by HPMC swelling. The optimum formulation contains 12% HPMC, 15% Eudragit RS and 25% plasticisant.
