5-NITROINDAZOLE DERIVATIVES –
A NEUROPROTECTIVE ALTERNATIVE
ON RAT ISCHEMIC BRAIN.

Abstract:
The aim of present study is to test in vivo neuroprotective activity of nine new synthetized 1-(piperazinyl-alkyl)-3-methyl-5-nitro-1H-indazole derivatives. The experiment was performed on 50 male adult Wistar rats divided into one control and nine working groups, each of five animals. The groups 1-9 were treated five days with 15 mg/kg/day orally with one of the N1-substituted-5-nitroindazole derivatives. Ischemic brain model was realized on all animals, occluding middle cerebral artery by our original technique. After euthanasia, the brains were removed and the cortical infarctus area measured using KS-300 Zeiss software. The neuroprotective action of the new synthesized compounds, estimated by the penumbra of cortical infarctus area (C.I.A.) reduction, demonstrated great differences. The most active derivatives (III, VIII) were selected for the development of future neuroprotective strategies involving neuronal NO-synthase inhibitors.
