Abstract:

The aim of this study was to analyze the mechanism of pharmacokinetic interaction between fluoxetine and metoclopramide. The clinical trial was in two periods and consisted in administration of a single dose of metoclopramide without or after 9 days treatment with fluoxetine. There were selected the pharmacokinetic parameters of metoclopramide which can be changed by fluoxetine and 8 pharmacokinetic models were built. After selection of the best model and data analysis, the results show the metabolic level of pharmacokinetic interaction between fluoxetine and metoclopramide, acting both on first pass effect (decreased by 50%) and on clearance (decreased by 21%).
